@davidasinclair @JoinLifespan You keep proclaiming you have the 1st FDA approved clinical trial of age reversal. That's false. The language by the FDA to @lifebiosciences was to clear a Phase 1 clinical trial testing ER-100 in people with certain optic-nerve diseases. David Sinclair (@davidasinclair) on the @joerogan podcast said that GLP-1s (such as Ozempic) have double sudden blindness rates It's difficult not to get irritated with this poor science communication. That alarm comes from 37 patients at one eye clinic. NAION is still about 1

Disappointing clinical study results continue to pile up for NR and NMN supplements In a 12-week double-blind, randomized, placebo-controlled pilot study of NR, there were no improvements in cognitive function (primary outcome), total CBF, or blood pressure (secondary outcomes)

@BradStanfieldMD Excellent! It is so important to publish negative studies with the same zest as positive ones. Great work with this one! 5.1K views · 154 likes · 6 reposts · 3 replies Open on X →

The bottom line: Exercise remains the single best intervention for preserving function in older adults. Full stop. I've always advocated that Rapamycin should NOT be used outside of FDA-approved indications and clinical trials Paper: https://t.co/c3B4Oq0fNz 11.8K views · 159 likes · 20 reposts · 13 replies Open on X →

I crowdfunded this trial with the help of @LifespanNews and @vita_dao. No pharma money. No conflicts with the drug. I genuinely hoped this would work. But science doesn't care about our hopes, and negative results matter just as much. 6.4K views · 147 likes · 4 reposts · 7 replies Open on X →

Important caveats: 1) This was 13 weeks. Longer trials may show different results 2) Only one dose/schedule tested. Less frequent dosing (every 3-6 weeks) might allow full drug clearance between exercise sessions 3) 40 participants. Why didn't the cycling hypothesis work? Leading theory is a pharmacokinetic problem. Rapamycin's half-life is ~62 hours. Even dosing it the day after exercise, active drug levels persisted into the next training sessions, partially blocking mTOR when muscles needed it most. 4.3K views · 46 likes · 1 reposts · 3 replies Open on X →

This parallels what we see with metformin. Two studies have shown metformin (also an indirect mTOR inhibitor) blunts exercise adaptations in older adults. 4.3K views · 58 likes · 5 reposts · 2 replies Open on X →

In the per-protocol analysis (people who actually followed the programme), the placebo group gained ~3.4 more chair-stand reps than the Rapamycin group. p = 0.007. Cohen's d = -0.90. That's a large negative effect. 4.8K views · 42 likes · 0 reposts · 1 replies Open on X →

Safety: both groups had similar rates of people reporting side effects (85% each). But the Rapamycin group had far more total events: 99 vs 63. One participant was hospitalised with pneumonia after a single Rapamycin dose. 4.7K views · 37 likes · 1 reposts · 1 replies Open on X →

We timed the rapamycin dose 24 hours AFTER the last exercise session each week, specifically to avoid blocking the anabolic window. Both groups improved. But the placebo group improved more. 5.1K views · 45 likes · 0 reposts · 3 replies Open on X →

We ran the first double-blind, placebo-controlled trial combining weekly Rapamycin (Sirolimus) 6mg with exercise in older adults. 40 sedentary people aged 65-85. Both groups did the same 13-week at-home exercise programme. Half got Rapamycin, half got Placebo. 6.8K views · 60 likes · 3 reposts · 4 replies Open on X →

The idea: the "cycling hypothesis." Exercise activates mTOR → muscle growth. Rapamycin inhibits mTOR → autophagy and cellular cleanup. Alternate the two and you get the best of both worlds. At least, that was the theory. 5.9K views · 46 likes · 0 reposts · 3 replies Open on X →

Our Rapamycin & Exercise clinical trial has just been published! The topline result? Rapamycin didn't help. Instead, it may have made things worse. Here's what we found 🧵 https://t.co/vpy9bPrGDJ https://t.co/GC6974lExP
