Whenever crossover exceeds 30% (as in this case), OS becomes very challenging to demonstrate. Which usually implies that the sequence (using the new regimen in 1L vs 2L) does not impact long term outcomes. In these cases, using the most tolerated regimen upfront is reasonable. 9.5K views · 87 likes · 15 reposts · 2 replies Open on X →

In a world of 300-pages protocols and standardized metrics Let’s please leave the truck test as it is No methods. No p-values. Just one rule. If you can drive a truck through the Kaplan-Meier curves, it passed the truck test. 🚒 https://t.co/BiZjYj2jD5

@PTarantinoMD @RenoHemonc Yes it passes the " truck test " But can we standardize the size of the truck In this era of AI we can make the truck very small that everyone can pass it if needed 3K views · 3 likes · 0 reposts · 1 replies Open on X →

Truck test ✅ A particularly neat feature of this trial: it compared an unconjugated topo1 inhibitor (topotecan) vs a conjugated topo1 inhibitor, demonstrating the impact of the ADC delivery system 🎯 https://t.co/T7jUxIKFmI

Multiple strategies are being pursued to optimize ADCs safety. Dose optimization is currently key to balance risks/benefits of ADCs. In the future, innovative constructs, diagnostic tools, improved conjugation & patient selection (eg. PGx) are expected to lead to improved safety.

Each ADC component (mAb, linker, payload) plays a critical role in determining the toxicity profile & can be modulated to improve it. Most toxicities are off-target & similar to those of the payload. However, mAb-related on-target effects can also lead to peculiar toxicities. htt

ADCs enable an improved delivery of chemo, expanding its activity, but not necessarily decreasing its side effects. Why is that? How can we reduce the toxicities of ADCs? Glad to share our @NatRevClinOncol article on how to optimize the safety of ADC! 🧵 https://t.co/QcpeKTosCq
